A Method of Staining Thin Sections with Lead Hydroxide for Precipitate-free Sections

چکیده

Staining of thin sections results in greater contrast which facilitates focusing and the observation of fine structure. Lead hydroxide, as suggested by Watson (1), has become increasingly popular as a stain for electron microscopy. However, when lead hydroxide is exposed to the carbon dioxide of air, lead carbonate forms and is deposited on the sections as fine, needle-like crystals, large amorphous or polygonal precipitates, or small granules. In order to prevent the stain from coming in contact with air, Peachey (2) used a plastic, air-free syringe as a receptacle for the stain and as a mechanism for staining sections. The cover of the syringe was filled with a carbon dioxide trap consisting of sodium hydroxide, anhydrous calcium chloride, and cotton. Parsons and Darden (3) designed a

A Technique for the Simultaneous Dirt-free Lead Staining of Several Electron Microscope Grids of Thin Sections

The staining of thin sections of Osmium tetroxide (OsO4)-fixed biological materials with the lead hydroxide preparat ion of Watson (5) or with semisaturated solutions of lead subacetate (1) represents a considerable advance in electron microscope technique. The contrast of the image viewed in the electron microscope is so far enhanced that accurate focusing is now possible at appreciably higher...

A Simplified Method of Staining Thin Sections of Biological Material with Lead Hydroxide for Electron Microscopy

A method of staining thin sections with lead hydroxide which enhances both resolution and contrast was described by M. L. Watson (1958) and is now undoubtedly in routine use in many laboratories. It is the purpose of this report to present a simplification of this method which eliminates the lengthy procedure of preparing the lead hydroxide reagent and the necessity of maintaining a COrfree atm...

A Device for Staining Tissue Sections for Electron Microscopy

It has been the experience of this laboratory that the lead hydroxide stain of Watson (1) gives excellent results in increasing contrast in thin sections of tissue, but that the stain is inconvenient to use because on contact with air it forms an insoluble precipitate which contaminates both the stored solution and the sections. 1 Contamination introduced in transferring the solution from the s...

A Routine Technique for Double-staining Ultrathin Sections Using Uranyl and Lead Salts

For more than 2 years, a double-staining of sections of OsO 4-fixed and Epon-embedded tissues with uranyl acetate followed by lead hydroxide has been routine technique in this laboratory. It was found to give substantially higher contrast, thus permitting the use of thinner sections. It facilitated focusing and routinely provided clearer pictures of cell fine structure than could be obtained wi...

THIN WALLED STEEL SECTIONS’ FREE SHAPE OPTIMIZATION USING CHARGED SYSTEM SEARCH ALGORITHM

Graph theory based methods are powerful means for representing structural systems so that their geometry and topology can be understood clearly. The combination of graph theory based methods and some metaheuristics can offer effective solutions for complex engineering optimization problems. This paper presents a Charged System Search (CSS) algorithm for the free shape optimizations of thin-wall...
